Human Intestinal Organoids and Application in Cystic Fibrosis

This course will allow you to learn basic and advanced culturing techniques of human intestinal organoids. We will focus on functional measurement of fluid secretion by the forskolin-induced swelling assay, and its use in the field of cystic fibrosis research, diagnostics and personalized medicine. This course is comprised of meeting experts and patients, hands-on bench work, and demonstrations. At the end of the training participants will have basic knowledge of organoids and their applications and are able to culture human intestinal organoids as well as to perform FIS assay experiments.

The development of sustainable human intestinal organoid stem cell cultures has been a massive breakthrough in human stem cell biology. Since organoid technology was developed by the research group of Prof. dr. Hans Clevers many potential applications have been proposed in clinical and research areas like oncology, gastro-enterology, microbiology and many more. Based on the human intestinal organoid stem cell model a fluid secretion assay has been developed which is capable of measuring CFTR function and response to therapy at the individual level. The forskolin induced swelling (FIS) assay (developed by the research group of dr. Jeffrey Beekman) is a valuable tool in cystic fibrosis drug development and patient stratification and has shown its potential for clinical patient representation and personalized medicine for CF-subjects.

This intensive course is offered by the University Medical Center Utrecht and the Beekman research group. During the course  you will learn the basic and advanced techniques that are necessary for successful isolation and culturing of human rectal organoids. Next to basic information about human intestinal organoids, we will zoom in on clinical applications of the organoid model focusing on cystic fibrosis. The forskolin induced swelling assay will be explained through interactive demonstration and hands- on  experiments by the participants. The course combines lectures from experienced and relevant speakers, tutorials with hands-on experiments and demonstrations. At the end of the training participants will have basic knowledge about organoids and applications and are able to culture human intestinal organoids as well as to perform FIS assay experiments.

Course aim

In this interactive course at a Masters level, you will learn the basic theory and techniques to work with rectal organoids for CF screening. The course combines lectures, tutorials with hands-on experiments, demonstrations and tours.
